Immune function changes of the IDPN‐induced Tourette syndrome rat model

Abstract There may be immunologic alternations during Tourette syndrome (TS) development. This study aimed to determine the immune function changes in different aspects (spleen or thymus index, plasma cytokines, and T cell) an 3,3′‐iminodipropionitrile (IDPN)‐induced rat model of TS. Male Sprague‐Dawley rats were assigned control TS groups. The group received intraperitoneal infections normal saline (5 ml kg −1 day ), injected with IDPN (150 mg ). spleen indices calculated. expression anti‐inflammatory cytokines inflammatory TNF‐α, peripheral blood measured by ELISA Western blotting. proportion CD3+, CD4+, CD8+, Treg, Th1, Th2 cells determined fluorescence‐activated cell sorting analysis. After 1 week treatment, had decreased weights versus control. levels IL‐4, IL‐10, IL‐12, IFN‐γ, TNF‐α significantly increased, while no significant difference TGF‐β was found. Flow cytometry analysis demonstrated that reduced CD3+ CD4+ spleen, without any change CD8+ cells. Furthermore, ratio Treg (CD4+/CD25+/FoxP3+) rats; simultaneously, Th1 (CD4+/IFN‐γ+) (CD4+/IL4+) dramatically increased. Together, can trigger dysfunction through impairment matured Th cells, particular for subset.
